Introduction:

Chemotherapy is the most effective treatment for most cancer patients. Many patients do not respond to therapy, and die of their disease. The cancer associated anorexia-cachexia syndrome is observed in 80% of patients with advanced stage cancer, and it is a very powerful prognostic indicator of poor outcome and poor quality of life. Moreover, this syndrome is also one of the major obstacles in cancer chemotherapy. Ghrelin is an orexigenic hormone which has been proposed to prevent anorexia. Therefore, this experiment is designed to determine whether addition of the Ghrelin agonist growth hormone releasing peptide 2 (GHRP-2) to cytotoxic therapy with 5-fluorouracil (5-FU) prevents anorexia associated with chemotherapy in cancer cachectic mice. Moreover, the chronic effect of GHRP-2 on reduced appetite and survival in this animal model were also been examined.

Thirty-three BALB/c female tumor-bearing mice were randomized to receive a solution including a). placebo; b) GHRP-2; c). 5-FU; d). 5-FU + GHRP-2. Ten of them received placebo solution. Food intake and survival were checked.

GHRP-2 is a synthetic agonist of ghrelin, the newly-discovered gut peptide which binds secretagogue receptor.
